Build an FTIR and complementary-analysis plan for ingredient identity

Use infrared evidence with qualified references and complementary methods, without treating a spectrum match as an automatic chelation percentage.

The practical answer

FTIR can contribute to a magnesium glycinate identity assessment when the method, reference and acceptance criteria are appropriate. Combine it with tests addressing different questions, such as elemental content, water and specific unwanted components. Do not assign a quantitative chelation percentage from an unexplained spectrum overlay.

Define the decision before collecting spectra

Mid-infrared spectroscopy measures absorption associated with molecular vibrations. The USP chapter provides the method framework. A useful identity procedure must explain what features distinguish the intended material from plausible alternatives under the actual preparation and measurement conditions. [1]

Start with the question: are you confirming a known approved grade, comparing an unfamiliar material with that grade, or investigating a suspected additional component? Each task may require different controls. A colorful graph without sample identifiers and an explanation of the comparison is not a complete identity record.

Assemble a comparison package

Water content, physical form, sample handling and data processing can influence comparisons. An investigation should therefore consider preparation differences before attributing every spectral change to a change in chemical identity. Equally, a superficially similar spectrum is not a reason to ignore contradictory composition data.

  • Identify the test lot, reference material, reference qualification and relevant specification revision.
  • Record preparation, measurement mode and conditions sufficient for another laboratory to understand the comparison.
  • Use pre-agreed acceptance criteria rather than choosing favorable peaks after seeing the result.
  • Include relevant negative or challenge materials when validating that the method can distinguish the alternatives that matter.
  • Retain the original data, processing choices and analyst interpretation, not only a cropped screenshot.

Choose complementary measurements for their purpose

This is an example of an orthogonal strategy: different measurements contribute information that is not simply a repeated version of the same signal. The exact combination should be chosen with an analytical laboratory familiar with the material. There is no universal fixed test panel that proves every commercial claim.

Close the identity review

Write a short conclusion separating observations from interpretation. For example, state that a sample met an approved identity procedure and the specified elemental assay, rather than expanding that result into an unsupported absorption or clinical claim. If a quantitative chelation claim is important, require its own defined measurand, method validation and calculation.

Use that principle to agree the package before purchase, then review method or reference changes through change control. [2] A supplier’s confidential process details need not all be public, but the buyer still needs enough evidence to make the intended quality decision.
